Jackie Peyton (Edie Falco) arbeitet als äußerst kompetente und erfahrene Krankenschwester im All Saints-Krankenhaus in New York City. Im Krankenhaus angekommen, verschwindet ihr Ehering in der Tasche und Lehrling Zoey erwartet sie bereits mit ihrem Häschen-Muster besetzten, rosafarbenen Arbeitskittel. Ihre Behandlungsmethoden als Krankenschwester sind allerdings meistens etwas fragwürdig, so unterscheidet sie gerne mal zwischen guten und bösen Patienten und behandelt diese dann dementsprechend. Auch ihre Kollegen, egal ob Arzt oder Auszubildende, kriegen hin und wieder Jackies Zorn zu spüren. Ganz nach dem Motto "Make me good god but not yet! " fälscht sie Organspender-Ausweise, lässt Körperteile verschwinden und schafft so nach besten Wissen und Gewissen fragwürdige Gerechtigkeit. Neben ihrem alltäglichen Arbeitsstress muss sie zusätzlich auch noch mit sich selbst fertig werden. Wenn sie abends geschafft heimkommt, warten sehnsüchtig schon ihre zwei süßen Töchter und ein liebevoller Ehemann auf sie... doch ihre Medikamentensucht und die Affäre mit dem Krankenhausapotheker Eddie machen ihr Leben nicht
