Der Reporter Newton Piles ist in der Hoffnung auf spannende Stories für den Chicago Chronicle in das Städchen Deliverance, Montana geraten. Er erreicht das Westernnest eine Woche vor der Einfahrt der ersten Eisenbahn, wird aber enttäuscht, was spannende Geschichten angeht. Einige Zeit später fällt eine Horde Vampir-Cowboys in dem Ort ein, tötet die Einwohner und entführt die Kinder. Newton Piles wird verschont, da der Anführer der Bande will, dass er von den Ereignissen berichtet. Die Bande wird von Billy the Kid angeführt, und wartet ebenfalls auf die Eisenbahn, um eine Armee aus Cowboy-Vampiren zu gründen. Mit der will er das ganze Land erobern.

Doch die Daywalkerin Rayne durchkreuzt diese Pläne, indem sie sich Verbündete sucht und den blutsaugenden Bestien den Kampf ansagt. Die aus Transylvanien stammende Rayne besitzt alle Stärken der Vampire, aber keine ihrer Schwächen. Außerdem hat sie mit Kid noch eine jahrhundertealte Rechnung offen ...

Der zweite Teil von Uwe Bolls dreiteiliger Film-Adaption des Video-Games "BloodRayne". Auch das Spiel hat zwei Teile und war Inspiration für zahlreiche Filme und Comicbücher. 2010 erschien das Sequel "BloodRayne: The Third Reich", bei dem ebenfalls Uwe Boll die Regie führte. Die Hauptdarstellerin aus "BloodRayne 1" Kristanna Loken stand wegen ihres Vertrags für den Action-Streifen "Painkiller Jane" und anderer Verpflichtungen nicht mehr zur Verfügung, ihren Part spielte deshalb - wie auch im dritten Teil - Natassia Malthe. Zwei Jahre später, 2007, arbeitete Kristianna Loken erneut mit Regisseur Uwe Boll zusammen, wieder für eine Video-Game-Verfilmung: "Schwerter des Königs - Dungeon Siege". Starinfo Michael Paré: Er ist Uwe Bolls Muse: In fast allen seinen Filmen spielt Michael Paré eine Rolle. Nach einer Kochlehre machte er Karriere zunächst als Model und verdiente vor allem mit Werbung in Magazinen und japanischen Clips eine Menge Geld. In den frühen 80er Jahren dann ließ sich der 1959 geborene Amerikaner bei Schauspiel-Guru Uta Hagen ausbilden. In der Folge feierte er seine größten Erfolge, unter anderem mit der US-amerikanischen TV-Serie "The Greatest American Hero" und als Rockikone in "Eddie and the Cruisers". Ein breiteres Publikum kennt ihn außerdem aus dem von John Carpenter produzierten Science Fiction "Das Philadelphia Experiment" und aus dem Actionfilm "Straßen in Flammen" (beide 1984). Paré ist zum dritten Mal verheiratet, seit 1992 mit dem Ex-Model Mrjolein Booy, und hat ein Kind mit ihr.
